Temperature-Sensitive Mutants of Measles Virus
were isolated after mutagenesis by 5-azacytidine, 5-fluorouracil, or proflavine. The restricted replication of all mutants at 39 C was blocked subsequent to cell penetration and could not be attributed to heat inactivation of virus infectivity. Complementation analysis was made possible through the use of poly-L-ornithine. The members of one complementation group exhibited wild-type RNA synthes...
متن کامل[Isolation of temperature-sensitive mutants of measles virus].
Measles virus differs from other members of paramyxoviruses in its prolonged growth cycle1), its degree of involvement of the cell nucleus2), its lack of neuraminidase3) and the sedimentability of its RNA4). We have undertaken a study of mutants of this virus in the hope that the study might circumvent some of the problems encountered with temperaturesensitive (ts) mutants of other paramyxoviru...
متن کاملComplementation analysis of measles virus temperature-sensitive mutants.
Two sets of independently isolated measles virus temperature-sensitive mutants were quantitatively tested for complementation. Analysis of the nine possible combinations of representative mutants indicated that only one pair of mutants is noncomplementing. Thus, the measles virus mutants studied to date define five complementation groups.
متن کاملIsolation and characterization of Sendai virus temperature-sensitive mutants.
Ten temperature-sensitive mutants of Sendai virus, a paramyxovirus, were isolated and partially characterized. The mutants replicated in chicken embryo lung cells at 30 C, but not at 38 C; wild-type virus grew equally well at both temperatures. Complementation tests divided the mutants into seven groups. Six groups synthesized neither infectious virus nor RNA when incubated at 38 C from the beg...
متن کاملTemperature-Sensitive Mutants of Vesicular Stomatitis Virus
Different temperature-sensitive mutants of vesicular stomatitis virus have been characterized in terms of their ability to induce synthesis of viral ribonucleic acid (RNA) in BHK-21 cells at 39 C (the restrictive temperature for these mutants). Mutants belonging to complementation groups I and IV (and probably II) did not induce actinomycin-resistant RNA synthesis in infected cells incubated at...
